Diagnostic Biosystems INSM1 A-8 INSM1 (A-8)
INSM1 (insulinoma-associated protein 1), also known as zinc-finger protein IA-1, is a developmentally regulated zinc-finger transcription factor. It localizes to the nucleus and is expressed in embryonic tissues undergoing neuroendocrine differentiation. INSM1 is not expressed in normal adult tissues but it can be found highly expressed in neuroendocrine tumors. INSM1 contains five Cys2-His2-type zinc-finger DNA binding domains and a prohormone domain. INSM1 acts as a transcriptional repressor of the Neuro D promoter and recruits cyclin D1 as a corepressor. It plays an importantrole in neuroendocrine development and isrequired for normal differentiation of pancreatic endocrine cells. Inhibition of INSM1 results in decreased formation of glucagon and Insulin positive cells. The gene encoding INSM1 is directly regulated by Neurogenin 3 which binds chromatin in the INSM1 promoter region and induces transcription.

This antibody recognizes a 200 kD glycoprotein (gp200) localized in the brush border of the proximal renal tebules and the luminal surface of Bowman’s capsule. gp200 was expressed by 93% of primary and 84% of metastatic renal carcinomas. Gp200 glycoprotein is also localized in the breast lobules and ducts, parathyroid glands and epididymis.

Sal-like protein 4 (SALL4) is a zinc-finger transcription factor that serves as a master regulator of embryonic pluripotency and is involved in processes associated with stem cell activities. SALL4 expression in germ cells makes it a useful marker for germ cell tumors such as seminoma, embryonal carcinoma, yolk sac tumors and teratomas. SALL4 expression is also seen in the spermatogonia of normal testis.

Recognizes a protein of 48kDa, which is identified as Adipophilin. It belongs to the perilipin family, members of which coat intracellular lipid storage droplets. This protein is associated with the lipid globule surface membrane material, and maybe involved in development and maintenance of adipose tissue. However, it is not restricted to adipocytes as previously thought, but is found in a wide range of cultured cell lines, including fibroblasts, endothelial and epithelial cells, and tissues, such as lactating mammary gland, adrenal cortex, Sertoli and Leydig cells, and hepatocytes in alcoholic liver cirrhosis, suggesting that it may serve as a marker of lipid accumulation in diverse cell types and diseases.

Hepatoblastoma is the most common primary tumor of the liver in children. The use of specific hepatocyte markers and also of α fetoprotein or carcinoembryonic antigen are useful for the identification of normal and malignant fetal hepatocytes. This antibody recognizes an uncharacterized antigen present in both adults and fetal normal hepatocytes to produce a distinct granular cytoplasmic staining. This antibody stains majority of hepatocellular carcinomas.
